Manchester United keeper Henderson has withdrawn from the tournament with a hip issue that would “continue to limit his involvement in training”.
He will return to his club for further assessment and treatment.
Posting on Twitter, Henderson wrote: “I worked so hard to be in this position. I’m devastated to have to withdraw through injury.”
He added: “I want to wish my team-mates all the best for the up and coming games. Bring it home.”
Sheffield United’s Ramsdale, 23, has no senior England caps but was part of Gareth Southgate’s provisional 33-man squad before the tournament.
He will follow Covid-19 testing protocol before entering the England camp.
“While I’m delighted to be joining back up with England, my immediate thoughts are with Deano as I know how much it meant to him to be involved,” Ramsdale said.
“I have now been given the honour and to be involved in a major tournament is what dreams are made of. This is a special moment for me and my family.”
Ramsdale will be one of three goalkeepers in the squad, joining Everton’s Jordan Pickford and West Bromwich Albion’s Sam Johnstone.
England beat Croatia in their opening game and face Scotland in their second Group D match at Wembley on Friday (20:00 BST).
Manchester United left-back Luke Shaw is part of the England squad but feared he might have been forced to miss the tournament after suffering a ruptured ligament in his wrist towards the end of the season.
Instead, he is part of the 26-man party, and bidding to force his way into the line-up.
“The doc had a look at it [the wrist] and couldn’t see the problem where the main ligament was, so he had to send it to a specialist,” said Shaw.
“I was a little bit worried, because I thought: If it is the bad one, then I might have to have an operation or something.
“He didn’t say anything and I was a little bit panicky but then obviously the next morning I’ve got the results and it wasn’t as bad as they thought so I was fine.”
Southgate picked right-back Kieran Trippier to play at left-back against Croatia instead of Shaw and fellow left-footed defender Ben Chilwell.
“I was obviously disappointed, but Gareth has 11 players he needs to pick and all I can do is train well, train hard and see what Gareth does in the next game,” added Shaw.
“But that’s the only thing I can do, keep smiling, keep enjoying it and do whatever I can to help the team, whether I am playing or not.”
